Resumo: Há inúmeras lacunas no conhecimento sobre as estratégias utilizadas por espécies de Leishmania para sobreviver dentro de macrófagos, dentre elas, a sobrevivência do parasita no ambiente inóspito do fagolisossomo e a modulação da resposta imune mediada por esta célula. O principal objetivo do estudo proposto é analisar as moléculas induzidas na célula hospedeira em resposta à infecção pelo parasita e que estão associadas à inibição do macrófago. Particularmente, o maior interesse é estudar os fatores que modulam a expressão
de CD200, uma molécula induzida após infecção por amastigotas de L. (L.) amazonensis e que modula a resposta do macrófago via iNOS. Desta forma, o presente projeto pretende estudar as vias de sinalização estimuladas pelo parasita e o potencial papel de receptores do tipo Toll (TLRs) na expressão de CD200 no macrófago e células fagocíticas.
Abstract: There are numerous gaps in the knowledge about the strategies used by Leishmania species to survive within macrophages, among them, parasite survival in the harsh environment of the phagolysosome, and modulation of the immune response mediated by this cell. The main objective of the proposed study is to analyze the molecules induced in the host cell in response to parasite infection that are associated with inhibition of macrophages. Particularly, the main interest is to study the factors that modulate the expression of CD200, a molecule induced after infection with amastigotes of Leishmania (Leishmania) amazonensis that modulate macrophage response by an iNOS dependent pathway. Thus, the present project aims to study signaling pathways stimulated by the parasite and the potential role of Toll-like receptors (TLRs) in the expression of CD200 on macrophage and phagocytic cells.
